Koetschau and Dr. G. Schmitt. In 1971 both companies merged with "The Commercial Laboratory Helmut Kleim",which was established in 1872. Finally "The Chemical Laboratory Dr. Gilbert", founded 1883, was integrated in 1973. Until 1995 Schmitt und Fintelmann (Inlabco) GmbH was a division of the Commercial and Environmental Laboratories Dr. Fintelmann und Dr. Meyer. In 1996 a separate company was founded. The activities of this company focuses on harbour/port services and maritime shipping, such as supervision of loading and unloading operations as well as gasfreeness-inspections. Until 1995 Dr. Fintelmann und Dr. Meyer Handels- und Umweltschutzlaboratorien was registered as a civil company, thereafter in the legal form as a commercial partnership. WATENSOL in plastic buckets (abt. 11 - 12 kg) is calculated for the production of 10 m cleaning solution (dilution 1 : 1000) The tank materials resp. tank coatings known to us (Stainless Steel, Dimetcoat, Epoxy, Carbozink) will not be affected by the cleaning solution (0.1%) produced with WATENSOL . Mikroorganismen sind mikroskopisch bzw. submikroskopisch klein. So weisen Bakterien einen Durchmesser von 1-3 m m auf. Die praktische Bedeutung der Mikroorganismen erkennt der Laie zun chst nur an den Sch den, die sie bei Menschen, Tieren und Pflanzen verursachen. So stehen die Mikroorganismen in der Rangfolge von Ern hrungsrisiken an zweiter Stelle. Ihre Stellung als N tzlinge vor allem in der Lebensmittelindustrie berwiegt diese Einstufung als Sch dlinge jedoch bei weitem. Ihre Verwendung erstreckt sich von der Veredlung landwirtschaftlicher Prim rprodukte bis hin zur Katalyse schwierigster chemischer Reaktionsschritte. Zu den klassischen mikrobiellen Verfahren geh ren die Herstellung von Bier unter Einsatz von Hefen , die Produktion von Milchprodukten mit Hilfe von Milchs urebakterien sowie die Herstellung von Speiseessig mit Hilfe von Essigs urebakterien . In der medizinischen Therapie sowie der pharmazeutischen Industrie begann eine neue Epoche, als die Wirkung der Ausscheidungsprodukte, allen voran des Penicillins , von Schimmelpilzen in der Behandlung bakterieller Infektionskrankheiten entdeckt wurde. In der Lebensmittelmikrobiologie sind drei Arten der Untersuchung zu unterscheiden. Zum Einen erfolgt die quantitative Erfassung von lebensf higen und vermehrungsf higen Mikroorganismen (Kolonie-Bildende-Einheiten, kurz: KBE ) auf einem geeigneten, nicht selektiven N hrboden. Weiterhin lassen sich auf selektiven N hrmedien qualitativ spezifische Mikroorganismen nachweisen. Und zuletzt erfolgt eine qualitative und quantitative Bestimmung verschiedener spezieller Mikroorganismen, ebenfalls auf einem selektiven N hrboden. In unseren Laboratorien pr fen wir routinem ig verschiedene Lebensmittel nach einem Standardprogramm auf die mikrobielle Belastung. Die hierf r angewandten Verfahren beruhen einerseits auf Methoden der Amtlichen Sammlung von Untersuchungsverfahren nach 35 des Lebensmittel- und Bedarfsgegenst ndegesetzes ( LMBG ). Da jedoch diese Verfahren nicht ersch pfend f r alle am Markt befindliche Lebensmittel vorliegen, sind diverse Verfahren aufgrund von Erfahrung im Umgang mit den Untersuchungsmethoden und durch Abstimmung mit weiteren Untersuchungslaboratorien entstanden. Diese zus tzlichen Verfahren wurden durch die Deutsche Akkreditierungssystem Pr fwesen GMBH ( DAP ) akkreditiert. Die leistungsf hige chemische Analytik ist zwar in der Lage, die Ausgangssubstanzen und alle ihre Metaboliten zu erfassen. ber die Wirkung auf Organismen werden jedoch keine Informationen gewonnen. Allerdings ist die Zahl der zu analysierenden Stoffe so umfangreich, dass eine vollst ndige Erfassung sehr aufwendig und kostenintensiv wird und synergistische und antagonistische Effekte nicht ber cksichtigt werden k nnen. Die kotoxikologischen Tests geben Auskunft ber die Wirkung, ohne den Stoff und damit die potentielle Quelle zu ermitteln. Sie erfassen aber neben der Bioverf gbarkeit verschiedene Effekte wie Abbau der vorhandenen Schadstoffe, die Bildung von Metaboliten sowie Transportereignisse und erm glichen eine konkrete Gef hrdungsabsch tzung. Dabei hat sich gezeigt, dass eine allgemeine kotoxikologische Aussage mit nur einem Testsystem nicht m glich ist. Aus diesem Grunde ist eine Kombination von biologischen Testverfahren zur Beurteilung von Umweltproben vorzuziehen. Aufkl rung Aufkl rung von Sch den an Beschichtungssystemen (Coating) von Schiffen Der Begriff Beschichtung ist gem DIN 55945 der Oberbegriff f r eine oder mehrere in sich zusammenh ngende, aus Beschichtungsstoffen hergestellte Schichten auf einem Untergrund. Bei mehrschichtigen Beschichtungen spricht man auch von einem Beschichtungsaufbau bzw. einem Beschichtungssystem . Beschichtungsstoffe sind fl ssige, past se oder auch pulverf rmige Stoffe, die zu deckenden und festhaftenden berz gen werden. Die Applikation dieser Beschichtungen hat neben dem dekorativen Zweck je nach Anwendungsbereich vor allem eine konservierende Wirkung. Diese kann jedoch nur dann eintreten, wenn der applizierte Beschichtungsstoff oder das gesamte Beschichtungssystem selbst haltbar ist. Es gen gt hierbei nicht, da der Stoff bzw. das System in sich dauerhaft ist. Es darf zus tzlich auch keine Abl sung von dem zu sch tzenden Untergrund stattfinden. Dies macht deutlich, welchen Wert die Kenntnis ber das zu beschichtende Material und die zu erwartende Belastung f r die Auswahl eines geeigneten Beschichtungsstoffes hat. Weiterhin sind auch die optimale Vorbereitung des Untergrundes sowie die Art der Applikation von entscheidender Bedeutung f r die Schutzwirkung der Beschichtung. In vorhergehenden Informationsbrosch ren unserer Laboratorien stellten wir bereits unsere Leistungen bei der Aufkl rung sogenannter Coatingsch den im Bereich der Schifffahrt dar. H ufigste Erscheinungsformen waren hier die Abl sung des applizierten Beschichtungssystems vom Untergrund sowie Unvertr glichkeitsreaktionen innerhalb der Beschichtungen. Diese stellten sich u.a. in Form von Blasenbildungen und Abl sungen innerhalb des Beschichtungsaufbaus dar. Die Ursachen f r diese Sch den sind vielf ltiger Natur. So kommen neben Verarbeitungsfehlern w hrend der Applikation oftmals auch mangelhafte Auswahl der Beschichtungsmaterialien vor. Als Beispiel sei hier ein Fall angef hrt, wo innerhalb eines Beschichtungssystems auf einer zinkhaltigen Fertigungsbeschichtung ein Alkydharz appliziert wurde. Die Folge war das Abl sen der Beschichtung vom Untergrund, wobei die Fertigungsbeschichtung auf dem Untergrund verblieb. Durch verschiedene Untersuchungsverfahren wie z.B. Gaschromatographie konnten an den Grenzfl chen der Beschichtungen Zinkseifen nachgewiesen werden. Diese deuteten auf eine chemische Reaktion der betreffenden Beschichtungsmaterialien hin, deren Folge ein Adh sionsbruch gewesen ist. Unsere Fachkenntnisse werden jedoch nicht nur zur Aufkl rung bereits vorhandener Sch den in Anspruch genommen. So f hren wir auch Eignungstests von Beschichtungsmaterialien f r Tankinnenbeschichtungen bezogen auf verschiedene Ladungsg ter durch. Auf diese Weise kann der Einsatz ungeeigneter Stoffe vermieden werden, die zwar konservierende Wirkung auf den Untergrund aufweisen, aber nicht bedingungslos f r die unterschiedlichsten Ladungen einsetzbar sind. The main criterias of the cleaning method are as follows: Our Tankcleaning method , based on the special cleaning agent WATENSOL , operates with the normal tank-wash-system of the vessel. WATENSOL in plastic buckets (abt. 11 - 12 kg) is calculated for the production of 10 m cleaning solution (dilution 1 : 1000) The tank materials resp. tank coatings known to us (Stainless Steel, Dimetcoat, Epoxy, Carbozink) will not be affected by the cleaning solution (0.1%) produced with WATENSOL . For the tankcleaning a solution of only 0.1% of our cleaning agent WATENSOL in fresh water is necessary, this allows the cleaning of tanks as well as lines and pumps from previous cargoes such as: MINERAL OILS PARAFFINES VEGETABLE OILS CHEMICALS up to a standard that chemicals of high purity can be loaded, such as: CHLORINATED HYDROCARBONS TOLUENE XYLENE GLYCOL METHANOL WATENSOL is no "dangerous good" in the regulations of transport such as land, sea and air. According to our knowledge, the use of WATENSOL neither produces any problems regarding the construction material of the loading tanks/lines/pumps, nor causes any danger to human health and environment if used as per our manuals. WATENSOL is admitted bei IMO as agent for tankcleaning under MARPOL annex II. WATENSOL - TANKCLEANING ensures a considerable reduction of the amount of slops as well an important sawing of time. Angaben Das Vorhaben "Vorbereitende Untersuchung f r die Entwicklung eines Testkits den Feststoffkontakttest mit Arthrobacter globiformis " ist Teil des Verbundprojektes ERNTE (Erprobung und Vorbereitung einer praktischen Nutzung kotoxikologischer Testsysteme). Der Kontakttest mit Arthrobacter globiformis erlaubt eine direkte kotoxikologische Pr fung eines kontaminierten Feststoffes durch die Bestimmung der Enzymaktivit t. Das Testdesign ist durch den direkten Einsatz von Bodenpartikeln als kologisch realistisch einzustufen. Die Bakterien werden der Probe zugesetzt, so dass eine Anpassung an die Kontamination nicht m glich ist, wie sie bei standorteigenen Mikroorganismen vorkommen kann. Der Kontakttest kann in ein Untersuchungskonzept eingebunden werden, das unterschiedliche Expositionswege beachtet. Dem Boden wird Arthrobacter globiformis aus der log-Wachstumsphase zugesetzt. Nach einer Kontaktzeit zwischen Boden und Organismus von zwei Stunden wird die Dehydrogenaseaktivit t im Vergleich zu einer unkontaminierten Kontrolle bestimmt. Testkriterium ist die Dehydrogenase-aktivit t, bemessen als Resazurinumsatz. Boden + Wasser + Testbakterien Ansatz + Resazurin ( in Puffer) Kontaktzeit 2 Stunden berkopfmischer Zentrifugation Reaktionszeit 50 Minuten berkopfmischer Messung Resazurinabnahme Photometer 600 nm Die Untersuchungen des TV 3 haben zum Ziel, den Kontakttest f r die kotoxikologische Untersuchung von B den unter realen Bedingungen zu testen und so zu optimieren, dass ein Testkit f r den praktischen Einsatz in der Umweltanalytik in absehbarer Zeit zur Verf gung gestellt werden kann. Hierf r soll das bereits normierte Verfahren miniaturisiert werden. Mit der Durchf hrung in Mikrotiterplatten ist auch die Umstellung der Messung des Resazurinumsatzes auf die fluorometrische Messung der Resofurinzunahme verbunden. Zur weiteren Entwicklung eines Testkits soll die Bereitstellung der Testorganismen in Form von lyophylisierten Bakterien gepr ft werden. Untersuchungen Konkret werden folgende Inhalte bearbeitet: Untersuchung des Einflusses unterschiedlicher Bodeneigenschaften auf das Ergebnis des Kontakttestes Fluorometrische Messung der Produktzunahme Resofurin mit und ohne Abtrennung des Feststoffes Testdurchf hrung auf Mikrotiterplatten Einsatz von konservierten Bakterien Untersuchungen zum Einsatz von Feststoff als Verd nnungsmedium zur Herstellung von Verd nnungsreihen Filtrationseinrichtung 96-well Mikrotiterplatte Der Bakterienkontakttest mit A. globiformis - 10 Jahre von der Entwicklung bis zur Normierung Der Bakterienkontakttest wurde an der TU Hamburg Harburg im AB Umweltschutztechnik entwickelt, um eine schnelle Beurteilung kontaminierter B den zu erreichen. Dabei sollte ein Testorganismus dem Feststoff zugesetzt werden (Kontakt Bakterien-Feststoff), der als Messparameter eine Enzymaktivit t ber cksichtigt, um eine allgemeine Stoffwechselreaktion zu erfassen und bei dem keine Extraktionsmittel eingesetzt werden m ssen.
